The latest Spanish scholar's research on hydrogen analgesia

Neuropathic pain with hyperalgesia as the main manifestation often becomes a chronic disease accompanied by mood disorders. Clinical treatments for neuropathic pain are still unsatisfactory and often have significant side effects.

In recent years, molecular hydrogen (H2) has been studied in a variety of diseases for its protective effects and unproven related secondary effects. The four main mechanisms by which H2 exerts neuroprotective effects include inhibition of oxidative stress, inflammatory and apoptotic processes, and regulation of autophagy. H2 produces its antioxidant properties by neutralizing OH and ONOO radicals. The gas also increased the expression of the antioxidant enzymes SOD-1 and HO-1. H2 also inhibited the activation of microglia and the secretion of pro-inflammatory cytokines (IL-1β, IL-6, TNF-α), and blocked the NF-κB pathway [20]. In terms of anti-apoptotic properties, we observed that it increased the ratio of the apoptosis regulator Bcl-2/BAX and decreased the expression of p-Akt and glycogen synthase kinase-3β. Finally, some studies have further shown that H2 can regulate autophagy by reducing the number of autophagosomes and altering the expression of different proteins, such as reducing extracellular signal-regulated kinase phosphorylation.

Mounting evidence of H2's therapeutic effects, coupled with its ability to cross the blood-brain barrier and limited side effects, make this gas a very interesting new approach to treating several neurological disorders such as CNP. Thus, previous studies have demonstrated that prophylactic treatment with hydrogen-rich water (HRW) or saline (HRS) is antinociceptive in rodents with neuropathic pain induced by nerve injury or spinal cord ligation. In addition, the antidepressant effect of HRW has also been described in animals with mild chronic unpredictable stress. However, the possible analgesic effects of therapeutic administration of HRW and its effects on anxiety and depression-like behavior in mice with chronic sciatic nerve constriction (CCI)-induced neuropathic pain have not been reported.

Consider the protective role of hydrogen molecule (H2) in different diseases, including neurological diseases such as Alzheimer's disease, and the antidepressant activity in chronically stressed animals. This study investigated the effects of hydrogen-rich water (HRW) on chronic sciatic nerve compression (CCI)-induced neuropathic pain and accompanying affective disturbances in male mice. We also investigated possible pathways for the analgesic activity of hydrogen-rich water and the interaction between heme oxygenase 1 (HO-1) and H2 in neuropathic pain.

the result shows:
(i) Repeated use of hydrogen-rich water has an inhibitory effect on hyperalgesia and hyperalgesia caused by chronic sciatic nerve compression;
(ii) Anxiolytic and antidepressant effects of hydrogen-rich water in animals with neuropathic pain;
(iii) The contribution of antioxidant enzymes (HO-1 and NAD(P)H:quinone oxidoreductase 1) and ATP-sensitive potassium channels in the analgesic effect of hydrogen-rich water in neuropathic pain;
(iv) a positive interaction of HO-1 and the hydrogen system in inhibiting chronic sciatic nerve compression-induced neuropathy;
(v) Antioxidative, anti-nociceptive, anti-inflammatory and/or anti-apoptotic characteristics of hydrogen water treatment in the dorsal root ganglia and/or amygdala of mice with sciatic nerve injury.
This study demonstrates a novel protective effect of hydrogen gas and suggests that hydrogen water may be an interesting therapeutic strategy for chronic neuropathic pain and its associated mood disorders.
